Trans-ISRIB is the active trans-isomer of the Integrated Stress Response Inhibitor (ISRIB). It is a small molecule capable of crossing the barrière hémato-encéphalique and effectively inhibiting the cellular integrated stress response (ISR). The ISR is typically activated by various stressors such as ER stress, amino acid deprivation, and viral infections, which results in the phosphorylation of the eukaryotic initiation factor 2 alpha (eIF2α). This process suppresses global protein synthesis while selectively allowing the translation of stress-response mRNAs. By acting downstream of eIF2α phosphorylation, Trans-ISRIB restores normal protein synthesis and is evaluated in various models for its cognitive and memory enhancement potential.
